Once you pick it up, you wear it. Once you get the hook claw, you should always use it to kill grubs. They will die in one hit, and you will have almost no chance of losing them when they fall down a hole. If you are having trouble beating any of the bonus levels, you can replay bonus level from the main menu or you have a second controller laying around, you can do them much easier with 2 players.
